PDRN and Amino Acids: Complete Building Blocks for Skin Regeneration

PDRN (polydeoxyribonucleotide) is a polymer of deoxyribonucleotides derived from salmon DNA that activates adenosine A2A receptors on fibroblasts, stimulating these cells to proliferate and produce collagen. However, fibroblasts can only synthesize collagen if sufficient amino acid precursors are available — collagen is approximately 33% glycine, 10% proline, and 10% hydroxyproline, with the remaining composition including alanine, arginine, and other amino acids. When amino acids are applied topically alongside PDRN, they serve as immediate precursors for the protein synthesis that PDRN's fibroblast activation initiates, removing the potential bottleneck of substrate availability.

The hydrating benefits are equally complementary. Amino acids are key components of the skin's Natural Moisturizing Factor (NMF) — the hygroscopic blend of substances within corneocytes that maintains stratum corneum hydration. Serine, glycine, alanine, and proline together constitute over 40% of free amino acids in the NMF. PDRN independently boosts hydration by stimulating glycosaminoglycan production (including hyaluronic acid) in the dermis. Age-related collagen decline results from both decreased fibroblast activity (which PDRN corrects through A2A activation) and reduced amino acid availability in aging tissue (which topical amino acids supplement). Studies show that aging skin has lower concentrations of NMF amino acids, contributing to the dryness and impaired barrier function characteristic of mature skin. In Korean skincare, amino acid-rich formulations (often derived from silk, soy hydrolysates, or fermented sources) are frequently layered with PDRN serums.
